Screening of Microorganism Producing Chitosanase Using Souble Chitosan and Its Enzymatic Characterization

Abstract
We investigated soluble chitosans as substrates to screen microorganism producing chitosanase on agar-plate including trypan blue. We prepared LB (spelled out) agar-plates including 0.02% trypan blue and 0.5% soluble chitosans such as 8kDa chitosan, 43kDa chitosan, and 67kDa chitosan, respectively. And then Bacillus subtilis, has been known as a microorganism producing chitosanase, was cultivated on the each plate. As a result, the 67kDa and 43kDa chitosans were determined as the proper substrates because the enzyme activity zone by chitosanase from B. subtilis was conformed clearly and the growth of microorganism was not inhibited on the plate including both chitosans as substrate, respectively. However, the activity zone was not showed on the plate including 8kDa chitosan as substrate. When the agar plate including 43kDa chitosan was used to screen microorganism producing chitosanase, one strain was isolated from soil, due to its forming ability clear zone. In the analysis of the 16SrRNA, the strain was identified as Bacillus sp. and was named this strain as Bacillus sp, NC2, The chitosanase from Bacillus sp. NC-2 exhibited the highest activities at pH 5.0 and 50oC and showed the highest enzyme activity for 67 kDa chitosan among various chitosan substrates.
